Allergy is a major public health issue in developed countries and its incidence is growing in emerging countries. The gastrointestinal tract is the first site of food allergic sensitization, and symptoms of food allergies are known to occur at other mucosal sites. To understand how the gut environment regulates allergic sensitization and allergic responses at other mucosal sites, and to elucidate the role of intestinal epithelial cells and microbiome on allergic diseases, we investigated (1) the role of NF-¿B pathway in epithelial cells, (2) the consequences of chronic exposure to an environmental pollutant, and (3) the role of Paneth cells NF-¿B is a transcriptional factor related with immunological responses including inflammation, and lymphoid organogenesis. Absence of canonical NF-¿B pathway in the intestinal epithelial cells due to IKKß-deficiency did not affect the development of allergen-specific IgE response, but altered the effective phase of allergy and reduced allergic responses were observed following oral allergen challenge. We have identified an impaired production of CCL11 by intestinal epithelial cells from IKKß ¿IEC mice as the protective mechanism that prevented the development of allergic symptoms in these mice.
